What is collagen?
Collagen is a kind of biological macromolecule, the main ingredient of connective tissue, which is widely found in the bones, skin, hair and blood vessels of animals. It plays the role of support, maintenance and repair of the body.
The main components of collagen are amino acids. Collagen can produce collagen peptide and byproducts amino acids after hydrolysis.
Collagen peptide has been widely used in food, medicine, tissue engineering, cosmetics and other fields because of its good biocompatibility, biodegradability and bioactivity.
The extraction method of collagen
The extraction methods of collagen mainly include acid method, alkali method, salt method, hot water extraction method and enzymatic method. Because the chemical method may destroy the biological active substances in collagen and produce some harmful byproducts, enzymatic extraction is the most commonly used method. In order to obtain better extraction effect, it is necessary to combine enzymatic extraction with hot water extraction in the actual extraction process.
The production process of collagen
The main influencing factors of enzymatic hydrolysis were: temperature, pH, enzyme activity, enzymolysis time, enzyme dosage and so on. In order to ensure that the raw material can reach a higher level of enzymatic hydrolysis, the appropriate enzyme type and hydrolysis time should be selected according to different materials in the practical operation.
Enzymes for the production of collagen
At present, the commonly used enzymes for collagen extraction include alkaline protease, neutral protease, trypsin, pepsin, papain and flavor protease. The complex enzyme is better than the single enzyme in enzymatic hydrolysis; it can improve the degree of hydrolysis and get the polypeptide with relatively small molecular weight.
Application of Annzyme enzyme in collagen production
Annzyme enzyme MF105, MF102 is the complex enzyme developed for hydrolysis of collagen.
After pretreatment and degreasing, the bovine skin was boiled at high temperature for 1 to 3 hours, then hydrolyzed at 55-60 ℃ for 3-6 hours, then centrifuged, filtered and dried to obtain collagen peptide powder.
Data of bovine skin collagen enzymatic hydrolysis
Test result of collagen peptide powder:
1) Total protein content > 90%
2) Total nitrogen content > 15%
3) Yield > 90%
4) The proportion of collagen peptides with molecular weight less than 2000Da is more than 90%.
5) The proportion of small peptides with molecular weight 400-180Da was 45%.
