Application of Selenium-Enriched Yeast Protein Hydrolysate in Animal Cell Culture

Selenium-enriched yeast protein hydrolysate is a functional nutritional substrate produced through specific processes including enzymatic hydrolysis, separation, concentration, and drying of selenium-enriched yeast. Compared to traditional yeast protein hydrolysates, it is not only rich in basic nutrients such as amino acids, peptides, vitamins, and trace elements, but also contains high levels of bioavailable organic selenium, giving it unique nutritional fortification value in the field of cell culture.

Compared to inorganic selenium (such as sodium selenite), organic selenium offers significant advantages in cell culture. These advantages stem from its higher bioavailability, lower cytotoxicity, and its ability to more accurately mimic the natural biological environment, thereby supporting more stable and efficient cellular physiological activities.

Literature studies confirm that selenium plays a key role in cell culture: it is involved in maintaining normal cell metabolism and function, and promotes cell proliferation and differentiation. However, high concentrations of inorganic selenium (such as sodium selenite) are cytotoxic, so it is typically added at low concentrations (on the order of µg/L) in culture systems as a serum substitute. Based on this, we designed a series of comparative experiments to evaluate the efficacy of Selenium-Enriched Yeast Protein Hydrolysate CM09.


In a low-serum culture system (2% serum) for mouse myoblast stem cells (C2C12), we conducted comparative tests between sodium selenite and CM09. The experimental results clearly showed that a certain concentration of CM09 had a significant promoting effect on cell proliferation under low-serum conditions, and its efficacy was superior to that of the low-concentration sodium selenite experimental group (see Fig 1). Further analysis of the working concentration revealed that the addition concentration of CM09 as a serum substitute is on the order of g/L, which is much higher than that of sodium selenite (on the order of µg/L) (see Table 1). To examine the applicability of CM09 in a broader range of cell types, we tested it in chicken fibroblast cells (DF-1) under both full-serum (10%) and serum-free culture conditions. The experimental results confirmed that CM09 can significantly promote cell proliferation and effectively replace some serum functions, highlighting its practical value in reducing serum dependence.

In summary, Selenium-Enriched Yeast Protein Hydrolysate CM09, with its high organic selenium content (>2000 ppm), excellent biocompatibility, and rich nutritional composition, demonstrates significant application potential in animal cell culture. Experimental data clearly confirm that CM09, under low-serum or even serum-free conditions, has superior cell proliferation-promoting effects compared to traditional inorganic selenium (sodium selenite) in various cell lines (such as C2C12 and DF-1), and can effectively replace part of the serum. This provides an efficient, safe, and naturally organic selenium-rich nutritional supplementation strategy for optimizing cell culture systems, particularly in applications with high serum dependency or those pursuing serum-free culture.
