Plant-derived and yeast-derived protein hydrolysates are natural mixtures rich in proteins, peptides of varying molecular weights, trace elements, vitamins, lipids, cholesterol, functional peptides, nucleotides, amino acids, and other components. They are produced from plant proteins, microbial whole cells, and other raw materials through directed enzymatic hydrolysis. Protein hydrolysates provide essential nutrients such as amino acids, nucleotides, vitamins, and carbohydrates for the growth of animal cells and are widely used in the culture of suspension and adherent cells.

Angel Yeast has developed several protein hydrolysates with different nutritional characteristics for use in animal cell culture. Angel CM05 is a peptide-rich yeast protein hydrolysate with low osmotic pressure; Angel CM06 is a low-calcium and magnesium yeast protein hydrolysate with low calcium and magnesium ion content; Angel CM07 is a glutathione-enriched yeast protein hydrolysate with glutathione content exceeding 10%; Angel CM08 is a self-digested yeast protein hydrolysate with balanced nutrition; and Angel PU041 and PU048 are soy protein hydrolysates rich in carbohydrates and vitamins. Extensive experimental validation has shown that Angel protein hydrolysates CM05, CM06, CM07, CM08, PU041, and PU048 can promote stem cell proliferation, maintain stem cell status, and partially replace serum in cell culture media, applicable in the field of cultured meat.

We selected common C2C12 mouse myoblast stem cells as the primary cell line for evaluating stem cell applications. (Figure 1) On the basis of a complete medium with 10% FBS, we added certain concentrations of Angel protein hydrolysates CM05, CM06, CM07, CM08, PU041, and PU048. After 48 hours of culture, CCK8 assays were conducted, and the results showed a certain promoting effect on C2C12 cell proliferation. When serum concentration was reduced to 5% (Figure 2), the experimental groups with Angel protein hydrolysates CM05, CM06, CM07, CM08, PU041, and PU048 still showed a promoting effect on C2C12 cell proliferation, with CM07, CM08, PU041, and PU048 experimental groups even exceeding the proliferation effect of the 10% FBS control group. When serum concentration was further reduced to 2% (Figure 3), under low-serum conditions, the experimental groups with Angel protein hydrolysates CM05, CM06, CM07, CM08, PU041, and PU048 still exhibited significant promoting effects on C2C12 cell proliferation, with the CM07, CM08, and PU041 experimental groups still exceeding the proliferation effect of the 10% FBS control group.

Extensive experimental evidence suggests that Angel protein hydrolysates can be used in the field of stem cell culture and partially replace serum in culture media. The amino acids and vitamins in Angel protein hydrolysates can provide the nutrients necessary for cell growth and proliferation, promoting cell growth and proliferation. Growth factors and polysaccharides in Angel protein hydrolysates can activate cellular metabolic pathways and enhance cell metabolic activity.
