To meet the unique requirements of certain applications, Angel Yeast has developed a reagent-grade low-calcium and low-magnesium yeast extract, CM06. "Low-calcium & low-magnesium" refers to the reduced levels of divalent metal ions such as calcium and magnesium in yeast extract. Conventional yeast extracts typically contain calcium levels greater than 600 mg/kg and magnesium levels exceeding 800 mg/kg. In contrast, CM06 yeast extract undergoes a specialized enzymatic process and specific removal of calcium and magnesium ions, resulting in calcium levels below 300 mg/kg and magnesium levels below 500 mg/kg. This reduction in calcium and magnesium ions while retaining other nutritional factors sets CM06 yeast extract apart.
CM06 yeast extract offers the following features:
1. High clarity: 1% solution has turbidity values below 0.5 NTU, and a 30% solution exhibits clear transparency. Additionally, it is compatible with most phosphate buffers, and no precipitation occurs during sterilization.
2. Light color: 2% solution exhibits an OD450 of less than 0.45 (measured after autoclaving at 121°C for 20 minutes).
3. Low endotoxin content: The endotoxin content is less than 10 EU/g, meeting the requirements for cell culture.
4. Rich in free amino acids and growth factors: CM06 contains over 30% free amino acids and is abundant in various B-group vitamins and nucleotides, supporting microbial growth and expression.
5. Strong growth-promoting capabilities: Even at low inoculation levels (10-100 CFU), CM06 stimulates rapid microbial growth and biomass accumulation.
Calcium and magnesium ions are vital elements for microbial growth and metabolism. They play various key roles in cell functions, including cell wall synthesis, nucleic acid synthesis, and enzyme activity regulation. However, excessive calcium and magnesium ion levels can inhibit the growth of lactic acid bacteria and lactobacilli. In cell culture processes, high calcium and magnesium ion levels may introduce high osmotic pressure, leading to cell aggregation. In research applications, low-calcium and low-magnesium yeast extract can be used to develop customized growth media for studying microbial calcium and magnesium-dependent reactions and growth characteristics.
For instance, in Enterococcus faecalis culture experiments, CM06 demonstrates its unique advantages. As shown in the figure below, compared to other products, CM06 yields higher biomass and leads to a lower pH after cultivation, indicating that CM06 better promotes the growth and acid production of Enterococcus faecalis.
In animal cell culture, due to the lower calcium and magnesium ion content in CM06, it introduces less osmotic pressure under the same concentration. As shown in the figure below, CM06 has lower osmotic pressure compared to other products, resulting in a higher number of viable cells in SF-9 cell culture.
In conclusion, low-calcium and low-magnesium yeast extract CM06 performs exceptionally well in specific industries, meeting the unique requirements of low calcium and magnesium in the fields of microbiology and animal cell culture.
